Influenza A H5 Subtyping by PCR

General Information

Lab Name

Influenza A H5 Subtyping by PCR

Lab Code

FLUH5

Epic Name

Influenza A H5 Subtyping by PCR

Description

This is a multiplex real-time PCR assay that targets Influenza A subtype H5 viruses (Hemagglutinin gene, specific for H5 subtype) and Influenza A viruses (Matrix gene). Results are reported separately as 'Detected' or 'Not Detected' for both subtype H5 and Influenza A. The lower limit of detection (LoD), defined as the lowest viral concentration that produces a positive result in 95% of replicates, is 230 copies/mL for subtype H5 and 1800 copies/mL for Influenza A.

Note: this assay was specifically developed for H5 subtyping and is not intended for use as a general screening tool for Influenza A virus.

This test was developed and its performance characteristics determined by the University of Washington Department of Laboratory Medicine and Pathology. It has not been cleared or approved by the US Food and Drug Administration.

How to order for UW Medicine patients:

        1. Order as a LADDON if there is already a sample tested by the lab (non POCT) for influenza virus - this applies to ED patients and inpatients, as well as any outpatient samples that have been sent to the lab.
        2. Order as a LABUND and collect a new sample if prior testing positive for influenza A virus is on a point-of-care instrument outside of our main lab sites - this applies to our UWPC locations that are running the Abbott ID Now platform.

          For Influenza screening:

Respiratory Virus PCR Panel (no SARS-CoV-2) [REVSQF]

Influenza A, B and RSV, Rapid PCR [FABRMC]

Influenza A, B & RSV PCR Panel [FLURSV]

Rapid Influenza A and B PCR [FLUMC]

COVID/FLU/RSV Respiratory Panel, Rapid PCR [FABRCV]

SARS-CoV-2 and Influenza A&B Qual PCR [NCVFLU]

          Ordering & Collection

          Specimen Type

Nasopharyngeal swab, Oropharyngeal swab, Anterior Nares swab and Eye swab.

          Collection

Collect swab specimens according to standard technique using a polyester-, rayon-, or nylon-tipped swab. Immediately place the swab specimen into VTM/UTM, M4 or M6 media. Do not use calcium alginate swabs. Refrigerate (2-8°C) within 4 hours of collection.

          Handling Instructions

Transport specimens on ice (ice Pak), store at 2 to 8°C for up to 72 hours post collection. If there is a greater than 72 hours delay before testing of the specimen, freeze at -70°C or colder and ship on dry ice. Do not freeze/thaw.

          Quantity

          requested: 1.0 mL
          minimum: 0.75 mL

          Processing

Send sample to UW Virology Laboratory within 4 hours of collection. If sample cannot arrive within 4 hours of collection, store sample at 2-8°C (ice pack) for up to 72 hours of collection. If there is a greater than 72 hour delay before testing sample, freeze sample at -70°C or colder and ship on dry ice. If freezer and/or dry ice is not available, ship immediately at 2-8°C (ice pack). Samples stored at 2-8°C greater than 72 hours old may be rejected. Do not freeze/thaw.
